Zusammenfassung: Ziel der vorliegenden Arbeit war es, die molekularen Mechanismen der apoptotischen Kooperation zwischen dem Todesrezeptor TNF-R1 und den Nicht-Todesrezeptoren der TNF-Rezeptorfamilie, insbesondere dem TNF-R2, aufzuklären. Es wurde hier gezeigt, dass die Art der zellulären Antwort, die vom TNF-R1 ausgeht, stark von der Dauer der TNF-Stimulation abhängen kann. So reicht ein TNF-Puls aus um den Transkriptionsfaktor NF-kB zu aktivieren, ist aber nicht hinreichend, um die apoptotische Zellmaschinerie in Gang zu setzen. Die Ko- und Vorstimulation des TNF-R2 konnten die TNF-vermittelte Apoptose-Induktion erhöhen und beschleunigen. Die Verstärkung der Apoptose korrelierte dabei mit der Zeitkinetik der TNF-R2-induzierten TRAF2-Depletion. Eine TNF-R2-Stimulation von 6 Stunden führte zu einer fast vollständigen Depletion von TRAF2 aus dem Zytoplasma, verstärkte den TNF-R1-vermittelten Zelltod dramatisch und inhibierte gleichzeitig die TNF-R1-induzierte NF-kB-Aktivierung zu 90%. Wurden die beiden TNF-Rezeptoren gleichzeitig aktiviert, so konnte noch immer eine beträchtliche Verstärkung der TNF-R1-induzierter Apoptose beobachtet werden, während die TNF-R1-vermittelte NF-kB-Aktivierung unbeeinflusst blieb. Mit Hilfe von der konfokalen Laserscanning-Mikroskopie konnte nachgewiesen werden, dass die Stimulation der Nicht-Todesrezeptoren CD40 und TNF-R2 zur TRAF2-abhängigen Rekrutierung der anti-apoptotischen Moleküle cIAP1 und cIAP2 an die Rezeptoren führte. Dies zeigt, dass der TNF-R1 und die Nicht-Todesrezeptoren der TNF-Rezeptorfamilie nicht nur um TRAF2 sondern auch um die mit TRAF2 assoziierten Schutzproteine kompetieren können. Diese Kompetition bildet den Kern des Modells zur apoptotischen Kooperation der beiden TNF-Rezeptoren. Demnach kann die Stimulation des TNF-R2 die TNF-R1-induzierte Apoptose auf zwei Ebenen verstärken: über die Inhibierung der NF-kB-abhängigen Induktion von Schutzproteinen und durch die Inhibition ihrer Schutzwirkung auf der post-transkriptionellen Ebene durch Kompetition. In beiden Fällen wird die Ausbildung eines Caspase-8-aktivierenden TNF-R1-Signalkomplexes ermöglicht. In vivo werden die Rezeptoren der TNF-Rezeptorfamilie auch durch membranständige Liganden aktiviert. Daher wurden im zweiten Abschnitt dieser Arbeit die Interaktionen zwischen TNF-R2, CD40 und ihren membranständigen Liganden analysiert. Unter Anwendung der Laser-Scanning-Mikroskopie und von Kokulturen konnte demonstriert werden, dass in Zell-Zell-Kontakten die Rezeptoren der TNF-Rezeptorfamilie mit ihren membranständigen Liganden Superaggregate bilden. Diese Aggregate entstehen innerhalb von 30 Minuten nach dem Zusammentreffen zweier Zellen, von denen eine den Rezeptor und die andere den membranständigen Ligand exprimiert. Die Rezeptor-Ligand-Superaggregate bleiben über mehrere Stunden stabil, wobei sich ihre Form und Lage stets verändern. Im Weiterem wurde beobachtet, dass die Rezeptor-Ligand-Superaggregate signalfähige Rezeptor-Komplexe enthalten. So wurde z.B. TRAF2 in die TNF-R2/memTNF-Aggregate rekrutiert. Wurde der CD40 mit einem löslichen CD40-Ligand stimuliert, so wurde die Internalisierung und möglicherweise der Abbau des Rezeptors beobachtet, nicht jedoch nach der Stimulation mit dem membranständigen Ligand. Dies deutet darauf hin, dass sich die Qualität der Bindung zwischen dem Rezeptor und seinem membranständigen Liganden wesentlich von der Interaktion des Rezeptors mit den löslichen Reagenzien unterscheidet.
The tumor necrosis factor (TNF) receptor family includes two groups of proteins: the death domain receptors and non-death domain receptors. TNF-R1, in contrast to TNF-R2, contains a death domain and is capable to induce apoptosis. Stimulation of TNF-R2 selectively enhances apoptosis induced by TNF-R1. The aim of this work was to explain the molecular mechanisms of this apoptotic crosstalk between TNF-R1 and non-death domain receptors of TNF receptor family. In response to TNF, TNF-R1 can activate different signaling pathways, leading either to induction of cell death or to activation of nuclear factors NF-kB and c-jun. Here it has been shown that TNF-R1-induced responses depend on the duration of TNF stimulation. A TNF pulse is sufficient for the activation of NF-kB, but it is insufficient for the induction of apoptosis. Co- and prestimulation of TNF-R2 effectively enhances TNF-R1-induced cell death. This selective enhancement of TNF-R1-induced apoptosis correlates with the kinetics of TNF-R2-induced TRAF2 depletion from the cytoplasm. Selective prestimulation of TNF-R2 for six hours results in the near to total depletion of TRAF2 from the cytoplasm. As a consequence, the induction of NF-kB-dependent anti-apoptotic factors was inhibited up to 90% and the cells were dramatically sensitized for TNF-R1-induced apoptosis. When both TNF receptors were stimulated simultaneously, TNF-R1-induced NF-kB activation remained unaffected but TNF-R1-induced cell death was still significantly enhanced. Compared with FasL-induced apoptosis, TNF-R1-induced activation of caspase-8 was clearly weaker and delayed. Costimulation or prestimulation of TNF-R2 enhances TNF-R1- mediated caspase-8 processing. Life cell imaging and confocal microscopy revealed that TNF-R2 and CD40 recruited the anti-apoptotic factors cIAP1 and cIAP2 in a TRAF2-dependent manner. This indicates that TNF-R2 and CD40 compete with TNF-R1 for the recruitment of TRAF2 and anti-apoptotic proteins. This competition plays a major role in the postulated model of the apoptotic crosstalk between TNF-R1 and non-death domain receptors of TNF receptor family, that interact with TRAF2 (Fig. 25). Exclusive stimulation of TNF-R1 with soluble TNF immediately activates the NF-kB-pathway leading to synthesis of NF-kB-dependent genes. The already existing and newly synthesized anti-apoptotic factors bind to TRAF2 and become recruited into TNF-R1 signaling complex, thereby preventing efficient activation of caspase-8. TNF-R2 prestimulation for several hours leads to TRAF2 depletion from cytoplasm and this has two consequences for TNF-R1 signaling. First, the activation of NF-kB is inhibited and therefore the synthesis of NF-kB-dependent genes is reduced. Second, low levels of pre-existing or still reduced levels of induced anti-apoptotic factors cannot work properly because they need TRAF2 support to be recruited to TNF-R1. Reduced NF-kB activation and impaired formation of TRAF2/TRAF1/cIAP1/cIAP2 complexes promotes the formation of apoptotic TRADD/FADD/Caspase-8 signaling complex. Caspase-8 becomes efficiently activated and apoptotic program is triggered. The situation changes again when both TNF receptors are stimulated simultaneously. TNF-R1-induced NF-kB activation is not inhibited under these conditions, because this process is clearly faster compared to TRAF2 depletion. TNF-R1 activation leads to normal production of anti-apoptotic factors. However, the biosynthesis of these factors needs time and during this time TRAF2 can be depleted/degradated by TNF-R2. Although anti-apoptotic proteins are produced, they cannot work properly, because their action at the post-transcriptional level requires TRAF2. Formation of apoptotic TNF-R1 signaling complexes is possible. As a consequence, cells are sensitized for TNF-R1-induced apoptosis, but the enhancement of apoptosis compared with the response after prestimulation is significantly weaker. Using confocal laser scanning microscopy it has been shown that TNF-R2 and CD40 form large aggregates with their corresponding membrane-bound ligands in cell-cell contacts. Receptor/ligand aggregates develop within 30 minutes after the induction. Induction means that cells express either receptor or membrane-bound corresponding ligand are joined together. These receptor/ligand aggregates are stable for many hours, however, their form and location change continuously. It could be demonstrated that receptor/ligand aggregates contain signaling competent complexes.
