Item Open Access A timed off-switch for dynamic control of gene expression in Corynebacterium glutamicum(2021) Siebert, Daniel; Altenbuchner, Josef; Blombach, BastianDynamic control of gene expression mainly relies on inducible systems, which require supplementation of (costly) inducer molecules. In contrast, synthetic regulatory circuits, which allow the timed shutdown of gene expression, are rarely available and therefore represent highly attractive tools for metabolic engineering. To achieve this, we utilized the VanR/PvanABK* regulatory system of Corynebacterium glutamicum, which consists of the transcriptional repressor VanR and a modified promoter of the vanABK operon (PvanABK*). VanR activity is modulated by one of the phenolic compounds ferulic acid, vanillin or vanillic acid, which are co-metabolized with d-glucose. Thus, gene expression in the presence of d-glucose is turned off if one of the effector molecules is depleted from the medium. To dynamically control the expression of the aceE gene, encoding the E1 subunit of the pyruvate dehydrogenase complex that is essential for growth on d-glucose, we replaced the native promoter by vanR/PvanABK* yielding C. glutamicum ΔPaceE::vanR-PvanABK*. The biomass yield of this strain increased linearly with the supplemented amount of effector. After consumption of the phenolic compounds growth ceased, however, C. glutamicumΔPaceE::vanR-PvanABK* continued to utilize the residual d-glucose to produce significant amounts of pyruvate, l-alanine, and l-valine. Interestingly, equimolar concentrations of the three phenolic compounds resulted in different biomass yields; and with increasing effector concentration, the product spectrum shifted from pyruvate over l-alanine to l-valine. To further test the suitability of the VanR/PvanABK* system, we overexpressed the l-valine biosynthesis genes ilvBNCE in C. glutamicum ΔPaceE::vanR-PvanABK*, which resulted in efficient l-valine production with a yield of about 0.36 mol l-valine per mol d-glucose. These results demonstrate that the VanR/PvanABK* system is a valuable tool to control gene expression in C. glutamicum in a timed manner by the cheap and abundant phenolic compounds ferulic acid, vanillin, and vanillic acid.Item Open Access Strategien der organisatorischen Segmentierung(1994) Reiß, Michael; Höge, RobertUntersuchungen zur Fertigungssegmentierung zeigen, daß sich neben anderen positiven Effekten die Herstellkosten um durchschnittlich zehn Prozent senken lassen. An solchen Erfolgsmeldungen wird heutzutage keiner achtlos vorübergehen. Alle Unternehmen sind gefordert, sich mit Segmentierung intensiv auseinanderzusetzen. Dabei umfaßt organisatorische Segmentierung mehr als nur die Fertigungssegmentierung. Angesichts der bunten Vielfalt von unterschiedlichen Segmentierungsformen müssen sich alle Praktiker über die zu verfolgende Strategie der optimalen Segmentierung im klaren werden.Item Open Access Resonant Raman scattering from superconducting single crystals of (BEDT-TTF)2I3(1993) Pokhodnia, Konstantin I.; Graja, Andrzej; Weger, Meir; Schweitzer, DieterA study of low energetic resonant Raman scattering of (BEDT-TTF)2I3 superconducors was performed. The vanishing of phonon bands accompanied by a decrease of the electronic background was observed below Tc. Das Sonderheft der WASSERWIRTSCHAFT enthält die Beiträge dieser Veranstaltung, welche vom Ministerium für Umwelt Baden-Württemberg gemeinsam mit dem Institut für Wasserbau der Universität Stuttgart ausgerichtet und vom Lehrstuhl für Technische Hydromechanik und Wasserbauliches Versuchswesen organisatorisch betreut wurde.Item Open Access Stand der Erkenntnisse über Laseranwendung(1989) Heisel, Uwe; Weiss, EdmundDie Laserindustrie mit ihrer Vielzahl von Anwendungsmöglichkeiten in der Produktion gilt als ein Wachstumsmarkt, dem hohe Zuwachsraten vorhergesagt werden. Trotz der zunehmenden Verbreitung des Lasereinsatzes zur Materialbearbeitung sind jedoch viele Anwendungsmöglichkeiten fertigungstechnisch noch nicht entwickelt. Dies trifft insbesondere auf die Holzbearbeitung zu, wo die Lasertechnik vergleichsweise erst am Beginn ihrer Entwicklungsmöglichkeiten steht. Im folgenden soll sowohl ein Überblick über die Möglichkeit und Chancen als auch über die Problemfelder der Laseranwendung in der Holzindustrie gegeben werden.Item Open Access Construction of a super-competent Bacillus subtilis 168 using the PmtlA-comKS inducible cassette(2015) Rahmer, Regine; Morabbi Heravi, Kambiz; Altenbuchner, JosefCompetence is a physiological state that enables Bacillus subtilis 168 to take up and internalize extracellular DNA. In practice, only a small subpopulation of B. subtilis 168 cells becomes competent when they enter stationary phase. In this study, we developed a new transformation method to improve the transformation efficiency of B. subtilis 168, specially in rich media. At first, different competence genes, namely comK, comS, and dprA, were alone or together integrated into the chromosome of B. subtilis 168 under control of mannitol-inducible PmtlA promoter. Overexpression of both comK and comS increased the transformation efficiency of B. subtilis REG19 with plasmid DNA by 6.7-fold compared to the wild type strain 168. This transformation efficiency reached its maximal level after 1.5 h of induction by mannitol. Besides, transformability of the REG19 cells was saturated in the presence of 100 ng dimeric plasmid or 3000 ng chromosomal DNA. Studying the influence of global regulators on the development of competence pointed out that important competence development factors, such as Spo0A, ComQXPA, and DegU, could be removed in REG19. On the other hand, efficient REG19 transformation remained highly dependent on the original copies of comK and comS regardless of the presence of PmtlA-comKS. Finally, novel plasmid-free strategies were used for transformation of REG19 based on Gibson assembly.Item Open Access The role of dimensionality and geometry in quench-induced nonequilibrium forces(2021) Nejad, Mehrana Raeisian; Khalilian, Hamidreza; Rohwer, Christian M.; Moghaddam, Ali GhorbanzadehWe present an analytical formalism, supported by numerical simulations, for studying forces that act on curved walls following temperature quenches of the surrounding ideal Brownian fluid. We show that, for curved surfaces, the post-quench forces initially evolve rapidly to an extremal value, whereafter they approach their steady state value algebraically in time. In contrast to the previously-studied case of flat boundaries (lines or planes), the algebraic decay for curved geometries depends on the dimension of the system. Specifically, steady-state values of the force are approached in time as t-d/2 in d-dimensional spherical (curved) geometries. For systems consisting of concentric circles or spheres, the exponent does not change for the force on the outer circle or sphere. However, the force exerted on the inner circles or sphere experiences an overshoot and, as a result, does not evolve to the steady state in a simple algebraic manner. The extremal value of the force also depends on the dimension of the system, and originates from curved boundaries and the fact that particles inside a sphere or circle are locally more confined, and diffuse less freely than particles outside the circle or sphere.Item Open Access Adjustable polystyrene nanoparticle templates for the production of mesoporous foams and ZnO inverse opals(2020) Abitaev, Karina; Qawasmi, Yaseen; Atanasova, Petia; Dargel, Carina; Bill, Joachim; Hellweg, Thomas; Sottmann, ThomasThe manifold applications of porous materials, such as in storage, separation, and catalysis, have led to an enormous interest in their cost-efficient preparation. A promising strategy to obtain porous materials with adjustable pore size and morphology is to use templates exhibiting the appropriate nanostructure. In this study, close-packed polystyrene (PS) nanoparticles, synthesized by emulsion polymerization, were used to produce porous PS and ZnO inverse opals. The size and distribution of the polystyrene nanoparticles, characterized by dynamic light scattering (DLS), small-angle neutron scattering (SANS), and scanning electron microscopy (SEM), were controlled via the concentration of sodium dodecyl sulfate (SDS). Systematic measurements of the water/styrene-interfacial tension show that the critical micelle concentration (CMC) of the ternary water–styrene–SDS system, which determines whether monodisperse or polydisperse PS particles are obtained, is considerably lower than that of the binary water–SDS system. The assemblies of close-packed PS nanoparticles obtained via drying were then studied by small-angle X-ray scattering (SAXS) and SEM. Both techniques prove that PS nanoparticles synthesized above the CMC result in a significantly unordered but denser packing of the particles. The polystyrene particles were subsequently used to produce porous polystyrene and ZnO inverse opals. While the former consists of micrometer-sized spherical pores surrounded by extended open-cellular regions of mesopores (Rpore ≈ 25 nm), the latter are made of ZnO-nanoparticles forming a structure of well-aligned interconnected pores.Item Open Access Lab-scale investigation of palm shell char as tar reforming catalyst(2020) Chen, Yen-Hau; Schmid, Max; Chang, Chia-Chi; Chang, Ching-Yuan; Scheffknecht, GünterThis research investigated the application of palm shell char as a catalyst for the catalytic steam reforming of tar after the sorption enhanced gasification (SEG) process. The catalytic activities of palm shell char and metal-supported palm shell char were tested in a simulated SEG derived syngas with tar model compounds (i.e., toluene and naphthalene) at a concentration of 10 g m-3 NTP. The results indicated that palm shell char had an experimentally excellent catalytic activity for tar reforming with toluene and naphthalene conversions of 0.8 in a short residence time of 0.17 s at 900 °C. A theoretical residence time to reach the complete naphthalene conversion was 1.2 s at 900 °C for palm shell char, demonstrating a promising activity similar to wood char and straw char, but better than CaO. It was also found that potassium and iron-loaded palm shell chars exhibited much better catalytic activity than palm shell char, while the parallel reaction of gasification of K-loaded palm shell char influenced the conversion with its drastic mass loss. Moreover, contrary to CaO, palm shell char presented relatively low selectivity to benzene, and its spontaneous gasification generated extra syngas. In summary, the present study demonstrated that the low-cost material, palm shell char, can successfully be used as the tar-reforming catalyst after SEG process.Item Open Access Multiphasic modelling and computation of metastatic lung-cancer cell proliferation and atrophy in brain tissue based on experimental data(2021) Ehlers, Wolfgang; Rehm, Markus; Schröder, Patrick; Stöhr, Daniela; Wagner, ArndtCancer is one of the most serious diseases for human beings, especially when metastases come into play. In the present article, the example of lung-cancer metastases in the brain is used to discuss the basic problem of cancer growth and atrophy as a result of both nutrients and medication. As the brain itself is a soft tissue that is saturated by blood and interstitial fluid, the biomechanical description of the problem is based on the Theory of Porous Media enhanced by the results of medication tests carried out in in-vitro experiments on cancer-cell cultures. Based on theoretical and experimental results, the consideration of proliferation, necrosis and apoptosis of metastatic cancer cells is included in the description by so-called mass-production terms added to the mass balances of the brain skeleton and the interstitial fluid. Furthermore, the mass interaction of nutrients and medical drugs between the solid and the interstitial fluid and its influence on proliferation, necrosis and apoptosis of cancer cells are considered. As a result, the overall model is appropriate for the description of brain tumour treatment combined with stress and deformation induced by cancer growth in the skull.
