Zusammenfassung: In der vorliegenden Arbeit wurde das Potential videomikroskopischer und bildanalytischer Methoden bei der Untersuchung des Zelltodes von eukaryotischen Zellpopulationen und einzelnen Zellen aufgezeigt. Zunächst wurde ein PC-gesteuertes Bildanalysesystem aufgebaut, mit dessen Hilfe die Bestimmung der Zellvitalität von CHO-Zellen (Chinese Hamster Ovary) in Fermentationsprozessen schnell und reproduzierbar durchgeführt werden kann. Die durchgeführten Untersuchungen zeigten, daß die Bestimmung lebender und toter Zellen bei der verwendeten CHO-Kultur mittels Vitalfärbung vorgenommen werden kann. Zusätzlich zu dem enormen Vorteil der raschen Auswertung vieler Proben ergab ein statistischer Vergleich der Zählergebnisse aus der manuellen und bildanalytischen Methode eindeutig eine bessere Reproduzierbarkeit der bildanalytischen Methode. Das aufgebaute Bildanalysesystem arbeitet mittels Programm-Makro automatisiert, so daß eine PC-gesteuerte on-line Anbindung an Prozeßleitsysteme von Fermentern möglich ist. Im zweiten Teil der Arbeit wurde das Videomikroskopiesystem für Fluoreszenzversuche entsprechend erweitert, um sehr spezifische Untersuchungen durchzuführen, welche eine weitere Aufklärung von Signalkaskaden im Ablauf des apoptotischen Zelltodes bringen sollten. Im Mittelpunkt dieser Untersuchungen stand die Lokalisierung und Quantifizierung einzelner intrazellulärer Signalmoleküle sowie die Charakterisierung mitochondrialer Funktionsänderungen. Dabei war es möglich, die Auswirkungen verschiedener Stimulantien und unterschiedlicher Mutanten auf die Bildung von membran-assoziierten Signalkomplexen unter physiologischen Bedingungen zu untersuchen und Aussagen über die zeitlichen Abläufe der Translokationsvorgänge zu machen. Unter Einsatz einer temperierten Zellkammer gelang die Quantifizierung des Signalmoleküls TRAF2 in diesen Signalkomplexen. Die gewonnenen Daten können in die Modellbildung eingebracht werden und so den Ausbau der entwickelten mathematischen Modelle für den ersten Teil der apoptotischen Signalkaskade, die Bindung von Rezeptor-Ligand und anschließende Bildung intrazellulärer membran-assoziierter Signalkomplexe, unterstützen.
This thesis presents the potential of video-microscopic and image-analytical methods for the investigation of the cell death of eucaryotic cell populations and individual cells. In order to determine the vitality of Chinese Hamster Ovary (CHO) cells in fermentation processes fast and quantitatively, an automated image analysis process is developed. The undertaken investigations show that alive and dead cells of the CHO culture taken can be determined by vitality coloring. The comparison of the manual determination method by microscopic counting of the cells and of the image analysis method shows next to the tremendous advantage of the fast analysis of many probes clearly a better reproducibility of the results when applying the image analysis. Since the assembled image analysis system is automated by program macros, a PC controlled on-line connection to process control systems of fermentors is possible. In the second part of this thesis, the image analysis system was extended, so that highly specific areas of interest of intracellular signal transduction processes during the apoptosis of individual cells could be investigated by fluorescence microscopic methods. Due to the high sensitivity of these methods very small amounts of fluorescencing molecules can be detected and quantitative determinations can be made. In the center of investigations is the localization and quantification of individual signal molecules during production of membrane-associated signal complexes under physiological conditions as well as the characterization of mitochondrial function changes. Thus, the investigations show the effects of several stimuli and different mutants on the generation of signal complexes as well as the dynamic behavior of transaction processes. For the proof of the translocation of the signal molecule TRAF2 from cytoplasm to the plasma membrane and the quantification of TRAF2 in the signal complexes, a temperature controlled cell chamber is constructed and applied. The resulting data can be used in mathematical models for the first part of the apoptopic signal cascade, namely the binding of the receptor to the ligand and the subsequent generation of the intracellular membrane-associated signal complex.
