Zusammenfassung: In dieser Arbeit wurde der Transkriptionsaktivator ManR des Bacillus subtilis Operons für den Mannose-Metabolismus näher charakterisiert. Er besteht aus einer N-terminalen DNA-Bindedomäne, zwei Phosphotransferasesystem (PTS)-Regula-tionsdomänen (PRDs) sowie einer EII_Bgl- und einer EIIA_Fru-ähnlichen Domäne. In Analogie zu anderen PRD-haltigen Regulatoren war eine Steuerung der Aktivität von ManR durch Phosphorylierung wahrscheinlich. Die ortsgerichtete Mutagenese von ManR wurde verwendet, um die Rolle seiner konservierten Aminosäure-Reste als potentielle Phosphorylierungsstellen zu untersuchen. Die Aktivität der ManR-Mutanten wurde in vivo über beta-Galaktosidase-Aktivitätstests sowie in vitro über Gel-mobilityshift Assays nachgewiesen. Für die in vitro Versuche mussten die isolierten ManR-Moleküle zuvor mit den PTS-Komponenten HPr, EI und PEP aktiviert werden. Mutationen in der PRD1 reduzierten die Aktivität von ManR, während Mutationen in der PRD2 diese vollständig aufhoben. Die Cys415Ala-(EIIB_Bgl) und die His570Ala-Mutationen (EIIA_Fru) riefen konstitutive Aktivitäten unterschiedlichen Ausmaßes hervor, wobei letztere den größeren Einfluss hatte. Im Wildtyp und in der Cys415Ala-Mutante verringerte die Anwesenheit der PTS-Transporterdomänen EIIBAMan die DNA-Bindeaktivität von ManR signifikant. Dies war jedoch nicht bei der His570Ala-Mutante der Fall. Durch den Austausch der konservierten Amino-säuren von ManR gegen Aspartat konnte keine Phosphomimikry beobachtet werden. Die unterschiedlichen Expressionsniveaus ausgehend von den ManR-kontrollierten Promotoren von manR (PmanR) und des manPA-yjdF Operons (PmanP) konnten der unterschiedlichen 5’-untranslatierten Region der mRNAs zugeordnet werden. Über DNase I Footprinting Experimente wurden palindromähnliche Sequenzen mit einer Länge von 44 bp als ManR-Bindestellen identifiziert. Des Weiteren wurden die Gleichgewichtsdissoziationskonstanten KD und die Dissoziationsraten kdiss von ManR bezüglich beider Promotorregionen bestimmt. Das Mannose-induzierbare Expressionssystem zur Produktion heterologer Proteine in B. subtilis führt zu relativ hohen Produktausbeuten, benötigt dafür jedoch hohe Mengen des teuren Induktors Mannose. Die Deletion des Gens für die Mannose-6-phosphat-Isomerase manA reduziert zwar den Induktorverbrauch, führt jedoch zur Sensitivität der Zellen gegenüber Mannose. Preisgünstiger, Mannose-haltiger Hefe-extrakt wurde als Induktoralternative getestet, zeigte jedoch nur eine geringe Induktionswirkung. Um die Produktausbeute bei gleichzeitiger Reduzierung der Induktorkosten zu steigern, wurde deshalb ein neuartiges, selbstinduzierendes Expressionssystem mit dem PTS-Transporter negativen Stamm B. subtilis TQ356 (spo0A3 delta_manP::ermC) entwickelt, bei dem Glucose die Induktion durch Katabolit-repression verhindert. Für optimale selbstinduzierende Bedingungen wurde eine Glucose-limitierende Prozessstrategie, nämlich ein Fed-Batch-Prozess, herangezogen: Die Selbstinduktion startet mit dem Beginn des Übergangs von der Batch- in die Zufütterungsphase, an dem Glucose limitierend wird, und bleibt während der gesamten Glucose-limitierten Zufütterungsphase erhalten. Dies führt im Vergleich zum Standardsystem zu einer Steigerung der Produktausbeute um fast das 3-fache. Der für das neue System verwendete, pUB110-basierte eGFP-Expressionsvektor (pMW168.1), der nach dem Rolling-Circle-Mechanismus repliziert, zeigte Instabilität bei der Segregation. Die Verwendung eines Replicons, welches nach dem Theta-Mechanismus repliziert, stabilisierte die Weitergabe an die Tochterzellen. Da mit diesem Replicon allerdings auch eine Reduzierung der Plasmidkopienzahl einher-ging, reduzierte sich entsprechend auch die Ausbeute und Produktivität des selbstinduzierenden Systems mit diesem Vektor. Selbst durch eine Erhöhung der Kopienzahl desselben Vektors wurden nicht die Werte erreicht, die mit pMW168.1 erzielt wurden. Die Anwendung der Alanin-Racemase als metabolischen Selektions-marker funktionierte zwar in Schüttelkolbenexperimenten, jedoch nicht bei der Anwendung in der Hochzelldichtefermentation. Die verwendeten Medien zeigten einen starken Einfluss auf die Expression von eGFP mit diesem System: Die Spurenelemente führten zu einem beschleunigten Wachstum, wodurch die Zellen länger in Glucose-Limitation waren und dement-sprechend mehr eGFP produzieren konnten. Der beobachtete negative Effekt der Casamino Acids auf die eGFP Expression wurde eingehend untersucht, führte jedoch zu keinem eindeutigen Resultat in Bezug auf eine bestimmte Komponente oder bestimmten Regulator.
In this study, the transcriptional activator ManR of the Bacillus subtilis mannose utilization operon was characterized. This protein is composed of an N-terminal DNA-binding domain, two phosphotransferase system (PTS) regulation domains (PRDs), an EIIB_Bgl- and an EIIA_Fru-like domain. In analogy to other PRD-containing regulators, stimulation of ManR activity by phosphorylation is proposed. Site-specific mutagenesis of ManR was used to reveal the role of its conserved amino acids as potential phosphorylation sites. This was investigated in vivo by beta-galactosidase activity tests and in vitro by mobility shift assays. For the in vitro experiments, ManR had to be first activated by the PTS components HPr, EI and PEP. Mutations in PRD1 lowered ManR activity, whereas mutations in PRD2 abolished it completely. The Cys415Ala (EIIB_Bgl) and the His570Ala mutations (EIIA_Fru) provoked constitutive activities to different degrees, whereas the latter had the greater influence. Addition of the PTS-transporter domains EIIBAMan reduced the binding capability significantly in a wild type and a Cys415Ala background, but had no effect on a His570Ala mutant. The conserved amino acids of ManR were also exchanged to aspartic acid. However, the intended phosphomimicry did not work with ManR. The different expression levels originating from the ManR-controlled promoters of manR (PmanR) and the manPA-yjdF operon (PmanP) could be ascribed to different 5´-un-translated mRNA regions. Sequences of 44 bps with a palindromic character were identified and confirmed as the ManR binding sites by DNase I footprinting. The binding properties of ManR, in particular the equilibrium dissociation constant KD and the dissociation rate kdiss, were determined for both promoter regions. The standard mannose-inducible expression system for heterologous protein production in Bacillus subtilis involves PmanP. It led to relatively high product yields but required large quantities of mannose to induce the reactions, thus rendering the system’s technical application with respect to the inducer costs too expensive. To reduce the amount of inducer, B. subtilis manA mutant strains were constructed, which, however, displayed sensitivity to mannose. Mannose containing yeast extract as a cheap inducer alternative was testet, but showed, however, only weak induction capabilities. With the aim of increasing product yields at the same time as decreasing production costs, a novel technically compliant and inducer-independent self-induction system with the PTS-transporter negative strain TQ356 (spo0A3 delta_manP::ermC) was developed, in which glucose prevents induction by carbon catabolite repression. To create optimal self-induction conditions, a glucose limited process strategy, namely a fed-batch process, was utilized: The initiation of self-induction at the beginning of the glucose-restricted transition phase between the batch- and feeding-phase of fermentation, and its maintenance throughout the entire glucose-limiting feeding-phase led to a nearly 3-fold increase of product yield compared to the standard system. The used pUB110-derived eGFP-expression vector (pMW168.1), which replicates after the rolling-circle mechanism, suffered from segregational instabilities. Exchange of the replicon to a more stable theta-replicating replicon raised its stability. Due to the accompanied reduction of the copy number, yield and productivity decreased with this system. Raising the copy number with respect to maintain the stability did not reach the levels gained with pMW168.1. Application of the alanine racemase as a metabolic selectable marker worked out well in shaking flask experiments, but failed in the application in a stirred-tank reactor. A strong influence of the used media on eGFP expression could be observed: Usage of the trace element solution raised the specific growth rate µ granting the cells more time in glucose limitation, which induced eGFP expression. The observed negative effect of the Casamino Acids on eGFP expression was studied, but could not be attributed to a certain component or regulator protein.
