Zusammenfassung: Die in Muttermilch mit bis zu 15 g l-1 enthaltenen humanen Milcholigosaccharide (HMOs) stellen eine bedeutende Gruppe bioaktiver Naturstoffe dar und zeigten in verschiedenen wissenschaftlichen Arbeiten zahlreiche positive Effekte für die Entwicklung von Säuglingen. Mit über 200 beschriebenen und über 100 strukturell charakterisierten Oligosacchariden weisen HMOs dabei eine große Strukturvielfalt auf. Untersuchungen der physiologischen Effekte von HMOs wurden in der Vergangenheit neben wenigen isolierten oder synthetisierten Reinstoffen häufig mit HMO-Gemischen durchgeführt, welche aus humaner Milch isoliert wurden. Der Grund hierfür ist, dass chemische Synthesen wegen der benötigten Schutzgruppenchemie aufwändig sind und enzymatische Synthesen mit Leloir-Glycosyltransferasen bzw. Glycosidasen kostspielige Nukleotid-aktivierte Zucker als Substrate benötigen bzw. geringe Produktausbeuten zeigen. Durch die Kombination hoher Regio- und Stereoselektivitäten von Leloir-Glycosyltransferasen mit einer kostengünstigen Bereitstellung von Nukleotid-aktivierten Zuckern waren in der Vergangenheit Ganzzellsynthesen einiger HMOs unter Verwendung von Plasmid-basierten Expressionen rekombinanter Gene beschrieben worden. Um die Synthese weiterer HMOs mittels Ganzzellsynthesen zu ermöglichen und dabei auf die Verwendung von Selektions-Systemen für Plasmid-tragende Zellen verzichten zu können, wurden in dieser Arbeit Plasmid-freie Stämme zur Synthese verschiedener HMOs konstruiert und die damit synthetisierten Oligosaccharide isoliert sowie analysiert. Die Stammkonstruktion erfolgte dabei durch chromosomale Integration von Expressionskassetten in Zuckerdegradations-Loci mittels homologer Rekombination und Selektion auf die Ortsspezifität der Integration mittels pH-Indikator Agarplatten. Unter Verwendung verschiedener Glycosyltransferase-Gene aus Helicobacter pylori, Escherichia coli und Neisseria meningitidis in Escherichia coli-Zellen, welche Nukleotid-aktivierte Zucker bereitstellen und Lactose aufnehmen, konnten so unter anderem die HMOs 2‘-Fucosyllactose (2‘-FL), 3-Fucosyllactose (3-FL), Lacto-N-tetraose (LNT), Lacto-N-neotetraose (LNnT), Lacto-N-fucopentaose I (LNFP I) und Lacto-N-difucohexaose II (LNDFH II) synthetisiert, isoliert und charakterisiert werden. Basierend auf den bereits vor dieser Arbeit am Institut für Mikrobiologie der Universität Stuttgart konstruierten E. coli Stämmen JM109 gwBC-F1 und JM109 gwBC F2-cat konnte eine Erhöhung der Produktivität von 2‘-FL durch die Erhöhung der chromosomal integrierten Kopien des Fucosyltransferase-Gens futC aufgezeigt werden. Außerdem konnte durch die zusätzliche Nutzung des Salvage Pathway für die Synthese von GDP-L-Fucose die Produktbildung weiter erhöht werden. Dabei wurden in Schüttelkolben-Kulturen mit Plasmid-freien Stämmen 2‘-FL-Titer von 1,06 ± 0,21 g l-1 bzw. 2,00 ± 0,04 g l-1 ohne bzw. mit Nutzung des Salvage Pathway erreicht. In einer darauffolgenden, Antibiotika-freien Zulaufkultivierung mit einem Endvolumen von 13,5 l und den Kohlenstoffquellen Glycerin und Lactose wurde ein 2‘-FL-Titer von 20,28 ± 0,83 g l-1 und somit etwa 270 g 2‘ FL mit einer Raum-Zeit-Ausbeute von 0,57 g l 1 h-1 produziert. In einem weiteren Teil dieser Arbeit konnte durch die Konstruktion des Stammes E. coli LJ-AYO-cat mit integrierten Genen einer Lactose-Permease, β1,3-N-Acetylglucosaminyl-transferase und β1,3-Galactosyltransferse erstmals die Synthese des HMO LNT in rekombinanten Mikroorganismen beschrieben werden. Dabei wurde unter Verwendung von Galactose als Kohlenstoffquelle im Vergleich zu Glycerin bzw. Glucose eine höhere intrazelluläre Konzentration des Donor-Substrats UDP-Galactose bestimmt, in Schüttelkolben 0,81 ± 0,01 g l-1 LNT synthetisiert und in einer Galactose-limitierten Zulaufkultivierung ein LNT-Titer von 12,72 ± 0,21 g l-1 erreicht. Bei einer Raum-Zeit-Ausbeute von 0,37 g l-1 h-1 wurden so 173 g des Tetrasaccharids LNT produziert. Durch die Ausstattung dieses LNT-synthetisierenden Stammes mit den Genen einer α1,2 bzw. α1,4-Fucosyltransferase und dem Salvage Pathway zur Synthese von GDP-L-Fucose wurden anschließend erstmalig rekombinante Ganzzellsynthesen fucosylierter Derivate von LNT beschrieben und dabei in Schüttelkolben-Kulturen 0,272 ± 0,006 g l-1 LNFP I bzw. 0,547 ± 0,004 g l-1 LNDFH II gebildet. Die Strukturen der gebildeten HMOs konnten nach der Isolierung jeweils mittles Massenspektrometrie und NMR-Spektroskopie aufgeklärt werden. Somit konnten in dieser Arbeit die Ganzzellsynthesen verschiedener HMOs mit Plasmid-freien Stämmen dargestellt, die Titer und Raum-Zeit-Ausbeuten in der Synthese von 2‘ FL erhöht und die Auswahl der mittels Ganzzellsynthesen herstellbaren HMOs um Typ I-HMO-Strukturen erweitert werden.
With up to 15 g l-1, human milk oligosaccharides (HMOs) represent an abundant and bioactive component of breastmilk whose positive effects on the well-being of infants were demonstrated in several scientific analyses. With more than 200 mentioned and over 100 structurally characterized oligosaccharides, HMOs show a great structural diversity. Due to the necessity of protection groups in chemical synthesis, low product yields in enzymatic synthesis with glycosidases and the need for costly nucleotide-activated sugars in the use of Leloir-glycosyltransferases, most HMOs are not available at reasonable prices. Besides few isolated or synthetic HMOs in pure form, previous analyses of the physiological effects of HMOs were therefore often conducted with mixtures of different HMOs that were isolated from human milk. By combining the regio- and stereoselectivity of Leloir-glycosyltransferases with the cost-efficient intracellular generation of nucleotide-activated sugars, previous publications could demonstrate whole-cell syntheses of some HMOs via plasmid-based expression of recombinant genes. To enable the synthesis of further HMOs using whole-cell systems and to avoid the need of selection-systems for plasmids, this work demonstrates the construction of plasmid-free strains for the synthesis of different HMOs, followed by product isolation and analysis. To do so, expression cassettes were chromosomally integrated into sugar-degradation loci by homologous recombination and selection for site-specificity of the integration on pH-indicator agar. By combining different glycosyltransferase-genes of Helicobacter pylori and Neisseria meningitidis in Escherichia coli cells, which provide nucleotide-activated sugars and incorporate, but do not to cleave lactose, the HMOs 2’-fucosyllactose (2’-FL), 3-fucosyllactose (3-FL), lacto-N-tetraose (LNT), lacto-N-neotetraose (LNnT), lacto-N-fucopentaose I (LNFP I) and lacto-N-difucohexaose II could be synthesized. Based on the strains E. coli JM109 gwBC-F1 and JM109 gwBC-F2-cat, previously constructed at the Institute of Microbiology, an increase in 2’-FL-productivity with an increased number of chromosomally integrated copies of the fucosyltransferase gene futC could be demonstrated. Furthermore, the additional usage of the salvage pathway for the generation GDP-L-fucose demonstrated an increase in product formation. In simple shake-flask cultivations, the plasmid-free strains produced 2’-FL-titers of 2,00 ± 0,04 g l-1 and 1,06 ± 0,21 g l-1 with or without the usage of the salvage pathway, respectively. In a subsequent antibiotic-free fed-batch cultivation with a final volume of 13,5 l and the carbon sources glycerol and lactose, a final 2’-FL-titer of 20,28 ± 0,83 g l-1 was reached. Thus, a total of about 270 g of 2’-FL could be synthesized with a space-time-yield of 0,57 g l-1 h-1. Furthermore, the first whole-cell synthesis of the HMO LNT using recombinant microorganisms could be demonstrated by chromosomally integrating the genes of a lactose-permease, a β1,3-N-acetylglucosaminyltransferase and a β1,3-galactosyl-transferase. By using galactose instead of glucose or glycerol as carbon source in shake-flask experiments, the intracellular concentrations of the donor-substrate UDP-galactose could be increased and a final LNT-titer of 0,81 ± 0,01 g l-1 could be reached with the strain E. coli LJ-AYO-cat. In a subsequent galactose-limited fed-batch cultivation a final LNT-titer of 12,72 ± 0,21 g l-1 could be reached with a space-time-yield of 0,37 g l-1 h-1, corresponding to a total amount of 173 g LNT. By equipping this LNT-producing strain with the genes of an α1,2- or α1,4-fucosyltransferase and the salvage pathway for the synthesis of GDP-L-fucose, the first whole cell synthesis of fucosylated derivatives of LNT in recombinant microorganisms could be described with final titers in shake-flasks of 0,272 ± 0,006 g l-1 LNFP I or 0,547 ± 0,004 g l-1 LNDFH II, respectively. Thus, in this work, several HMOs were successfully synthesized in whole-cell systems using plasmid-free E. coli strains. Furthermore, the previously described titers and space-time-yields in the synthesis of 2’-FL were significantly increased and the variety of HMOs available by whole-cell synthesis was expanded by Type I-HMO-structures.
