    ROSIE : RObust Sparse ensemble for outlIEr detection and gene selection in cancer omics data

    The extraction of novel information from omics data is a challenging task, in particular, since the number of features (e.g. genes) often far exceeds the number of samples. In such a setting, conventional parameter estimation leads to ill-posed optimization problems, and regularization may be required. In addition, outliers can largely impact classification accuracy. Here we introduce ROSIE, an ensemble classification approach, which combines three sparse and robust classification methods for outlier detection and feature selection and further performs a bootstrap-based validity check. Outliers of ROSIE are determined by the rank product test using outlier rankings of all three methods, and important features are selected as features commonly selected by all methods. We apply ROSIE to RNA-Seq data from The Cancer Genome Atlas (TCGA) to classify observations into Triple-Negative Breast Cancer (TNBC) and non-TNBC tissue samples. The pre-processed dataset consists of 16,600 genes and more than 1,000 samples. We demonstrate that ROSIE selects important features and outliers in a robust way. Identified outliers are concordant with the distribution of the commonly selected genes by the three methods, and results are in line with other independent studies. Furthermore, we discuss the association of some of the selected genes with the TNBC subtype in other investigations. In summary, ROSIE constitutes a robust and sparse procedure to identify outliers and important genes through binary classification. Our approach is ad hoc applicable to other datasets, fulfilling the overall goal of simultaneously identifying outliers and candidate disease biomarkers to the targeted in therapy research and personalized medicine frameworks.

    Alumina and zirconia-reinforced polyamide PA-12 composites for biomedical additive manufacturing

    This work aimed to prepare a composite with a polyamide (PA) matrix and surface-modified ZrO2 or Al2O3 to be used as ceramic fillers (CFs). Those composites contained 30 wt.% ceramic powder to 70 wt.% polymer. Possible applications for this type of composite include bioengineering applications especially in the fields of dental prosthetics and orthopaedics. The ceramic fillers were subjected to chemical surface modification with Piranha Solution and suspension in 10 M sodium hydroxide and Si3N4 to achieve the highest possible surface development and to introduce additional functional groups. This was to improve the bonding between the CFs and the polymer matrix. Both CFs were examined for particle size distribution (PSD), functional groups (FTIR), chemical composition (XPS), phase composition (XRD), and morphology and chemical composition (SEM/EDS). Filaments were created from the powders prepared in this way and were then used for 3D FDM printing. Samples were subjected to mechanical tests (tensility, hardness) and soaking tests in a high-pressure autoclave in artificial saliva for 14, 21, and 29 days.

    Lernen und üben wir das Richtige? : Kritische Erfolgsfaktoren der Bewältigung des Massenanfalls von Verletzten : Ergebnisse einer FMEA und einer Analyse von MANV-bezogenen Curricula

    Der Massenanfall von Verletzten (MANV) ist eine Ausnahmesituation für Rettungsdienst und andere Einsatzkräfte. Trotz niedriger Inzidenz müssen sich die Einsatzkräfte sowohl auf ärztlicher als auch auf nichtärztlicher Seite auf diesen Einsatzfall vorbereiten. In der vorliegenden Pilotstudie wurden anhand einer Fehlermöglichkeits- und -einflussanalyse (FMEA) die kritischen Einsatzfaktoren im MANV ermittelt und mit den Ausbildungscurricula verglichen. Die herangezogenen Curricula waren Notfallsanitäter:in, organisatorische:r Leiter:in, Konzept zur katastrophenmedizinischen Ausbildung im studentischen Unterricht an deutschen Hochschulen, Nationaler Kompetenz-basierter Lernzielkatalog Medizin (NKLM), Zusatz-Weiterbildung Notfallmedizin und Fortbildung zum:zur leitenden Notarzt:ärztin. Die Ergebnisse lassen vermuten, dass in der praktischen Ausbildung der MANV eine eher untergeordnete Rolle spielt. Weitere empirische Untersuchungen sind aus Sicht der Autoren notwendig.

    Numerical modeling of cutting characteristics during short hole drilling : modeling of kinetic characteristics

    Analyzing the cutting process characteristics opens up significant opportunities to improve various material machining processes. Numerical modeling is a well-established, powerful technique for determining various characteristics of cutting processes. The developed spatial finite element model of short hole drilling is used to determine the kinetic characteristics of the machining process, in particular, the components of cutting force and cutting power. To determine the component model parameters for the numerical model of drilling, the constitutive equation parameters, and the parameters of the contact interaction between the drill and the machined material on the example of AISI 1045 steel machining, the orthogonal cutting process was used. These parameters are determined using the inverse method. The DOE (Design of Experiment) sensitivity analysis was applied as a procedure for determining the component models parameters, which is realized by multiple simulations using the developed spatial FEM model of orthogonal cutting and the subsequent determination of generalized values of the required parameters by finding the intersection of the individual value sets of these parameters. The target values for the DOE analysis were experimentally determined kinetic characteristics of the orthogonal cutting process. The constitutive equation and contact interaction parameters were used to simulate the short hole drilling process. The comparison of experimentally determined and simulated values of the kinetic characteristics of the drilling process for a significant range of cutting speed and drill feed changes has established their satisfactory coincidence. The simulated value deviation from the corresponding measured characteristics in the whole range of cutting speed and drill feed variation did not exceed 23%.

    Digital factory transformation from a servitization perspective : fields of action for developing internal smart services

    In recent years, a complex set of dynamic developments driven by both the economy and the emergence of digital technologies has put pressure on manufacturing companies to adapt. The concept of servitization, i.e., the shift from a product-centric to a service-centric value creation logic, can help manufacturing companies stabilize their business in such volatile times. Existing academic literature investigates the potential and challenges of servitization and the associated development of data-based services, so-called smart services, with a view to external market performance. However, with the increasing use of digital technologies in manufacturing and the development of internal smart services based on them, we argue that the existing insights on external servitization are also of interest for internal transformation. In this paper, we identify key findings from service literature, apply them to digital factory transformation, and structure them into six fields of action along the dimensions of people, technology, and organization. As a result, recommendations for designing digital factory transformation in manufacturing companies are derived from the perspective of servitization and developing internal smart services.

    A nonsmooth generalized‐alpha method for mechanical systems with frictional contact

    In this article, the existing nonsmooth generalized‐α method for the simulation of mechanical systems with frictionless contacts, modeled as unilateral constraints, is extended to systems with frictional contacts. On that account, we complement the unilateral constraints with set‐valued Coulomb‐type friction laws. Moreover, we devise a set of benchmark systems, which can be used to validate numerical schemes for mechanical systems with frictional contacts. Finally, this set of benchmarks is used to numerically assert the properties striven for during the derivation of the presented scheme. Specifically, we show that the presented scheme can reproduce the dynamics of the frictional contact adequately and no numerical penetration of the contacting bodies arises - a big issue for most popular time‐stepping schemes such as the one of Moreau. Moreover, we demonstrate that the presented scheme performs well for multibody systems containing flexible parts and that it allows general parametrizations such as the use of unit quaternions for the rotation of rigid bodies.

    Statistical power analysis in reliability demonstration testing : the probability of test success

    Statistical power analyses are used in the design of experiments to determine the required number of specimens, and thus the expenditure, of a test. Commonly, when analyzing and planning life tests of technical products, only the confidence level is taken into account for assessing uncertainty. However, due to the sampling error, the confidence interval estimation varies from test to test; therefore, the number of specimens needed to yield a successful reliability demonstration cannot be derived by this. In this paper, a procedure is presented that facilitates the integration of statistical power analysis into reliability demonstration test planning. The Probability of Test Success is introduced as a metric in order to place the statistical power in the context of life test planning of technical products. It contains the information concerning the probability that a life test is capable of demonstrating a required lifetime, reliability, and confidence. In turn, it enables the assessment and comparison of various life test types, such as success run, non-censored, and censored life tests. The main results are four calculation methods for the Probability of Test Success for various test scenarios: a general method which is capable of dealing with all possible scenarios, a calculation method mimicking the actual test procedure, and two analytic approaches for failure-free and failure-based tests which make use of the central limit theorem and asymptotic properties of several statistics, and therefore simplify the effort involved in planning life tests. The calculation methods are compared and their respective advantages and disadvantages worked out; furthermore, the scenarios in which each method is to be preferred are illustrated. The applicability of the developed procedure for planning reliability demonstration tests using the Probability of Test Success is additionally illustrated by a case study.

    Comprehensive study of failure mechanisms of field-aged automotive lead batteries

    Modern vehicles have increasing safety requirements and a need for reliable low-voltage power supply in their on-board power supply systems. Understanding the causes and probabilities of failures in a 12 V power supply is crucial. Field analyses of aged and failed 12 V lead batteries can provide valuable insights regarding this topic. In a previous study, non-invasive electrical testing was used to objectively determine the reasons for failure and the lifetime of individual batteries. By identifying all of the potential failure mechanisms, the Latin hypercube sampling method was found to effectively reduce the required sample size. To ensure sufficient confidence in validating diagnostic algorithms and calculating time-dependent failure rates, all identified aging phenomena must be considered. This study presents a probability distribution of the failure mechanisms that occur in the field, as well as provides insights into potential opportunities, but it also challenges diagnostic approaches for current and future vehicles.

    Farbkodierte objektangepasste Streifenprojektion für die schnelle 2D- und 3D-Qualitätsprüfung

    Die Kontrolle von Werkstücken hinsichtlich ihrer dreidimensionalen Geometrie und ihrer Oberflächencharakteristik ist von entscheidender Bedeutung für die automatisierte industrielle Produktion. Vorgestellt wird ein schnelles und robustes Einzelbildverfahren das es gestattet, gleichzeitig die Topographie zu kontrollieren und ein Videobild des Werkstückes zu generieren. Hierzu wird eine objektangepasste Streifenprojektion mit einer farbkodierten Maske verwendet. Zur Detektion können preiswerte Einzelchip Farbkameras zum Einsatz kommen. Das Verfahren erzielt eine vergleichsweise gute 3D-Auflösung von 1/4000 bei einer hohen lateralen 2D- und 3D-Auflösung und ist weitgehend unempfindlich gegen lokale Verschmutzungen und Variation der Beleuchtung.

    Cooperative search by combining simulated and real robots in a swarm under the view of multibody system dynamics

    This paper presents a new approach for cooperative search of a robot swarm. After modeling the robot, the mechanical Particle Swarm Optimization method is conducted based on physical robot properties. Benefiting from the effective localization and navigation by sensor data fusion, a mixed robot swarm which contains both simulated and real robots is then successfully used for searching a target cooperatively. With the promising results from experiments based on different scenarios, the feasibility, the interaction of real and simulated robots, the fault tolerance, and also the scalability of the proposed method are investigated.
