Zusammenfassung: Das Verbreitungsgebiet des Europäischen Feldhamsters (Cricetus cricetus) erstreckt sich über die gemäßigten Zonen Eurasiens und ist durch dramatische saisonale Veränderungen im Klima, der Vergetation und der Futterverfügbarkeit gekennzeichnet. In Anpassung an diese saisonalen Wechsel zeigen Europäische Feldhamster ausgeprägte saisonale Veränderungen in ihrer Physiologie und im Verhalten. Die Reproduktionszeit ist auf das Frühjahr und den frühen Sommer beschränkt, dagegen werden die harten Bedingungen des Winters durch Winterschlaf umgangen. Außerdem steigt das Körpergewicht in der ersten Jahreshälfte und sinkt nach der Sommersonnenwende. Diese saisonalen Veränderungen haben eine starke endogene Komponente, denn sie bestehen sogar unter konstanten Bedingungen (Canguilhem, 1989), d.h. sie spiegeln wahrscheinlich einen echten circannualen Rhythmus wider. Überdies werden saisonale Veränderungen in zwei circadianen Funktionen, nämlich dem Aktivitätsmuster und der Melatoninsekretion beobachtet. Während des größten Teils des Jahres ist der Aktivitätsrhythmus schwach oder arhythmisch. Hohe Aktivitätslevel und ein präzises -muster werden nur während zwei bis drei Monaten um die Sommersonnenwende beobachtet (Wollnik et al., 1991). Dagegen ist ein Tag-Nacht-Rhythmus in der Melatoninsekretion nur während mittlerer und kurzer Photoperioden feststellbar (Vivien-Roels et al., 1992). Normalerweise unterscheidet sich die Periodenlänge der endogenen circannualen Rhythmen vom annualen Zyklus der Umwelt (weshalb sie circa-annual genannt werden). Deshalb muss die circannuale Uhr wenigstens einmal pro Jahr auf die Oszillation der Umwelt synchronisiert werden. Es ist allgemein anerkannt, dass die jährlichen Veränderungen in der Tageslichtlänge (Photoperiode) das wichtigste Umweltsignal dafür liefern. Dieses Dissertation beschäftigt sich mit der Frage, wie der photoperiodische Synchronisationsmechanismus bei Eurooäischen Feldhamstern funktioniert. Die Synchronisation von jährlichen Rhythmen beim Europäishcen Feldhamster ist das Resultat einer Interaktion zwischen saisonalen Veränderungen in der Photoperiode und saisonalen Veränderungen in der Empfindlichkeit gegenüber der Photoperiode. Eine sensitive Phase für kurze Photoperioden dauert von Mitte Mai bis Mitte Juli, wenn eine Abnahme in der Tageslichtlänge zur Gonadenregression und Abnahme des Körpergewichts führt (Saboureau et al., 1999). Im ersten Teil der vorliegenden Arbeit wird untersucht, ob es eine weitere sensitive Phase für den gegenteiligen Wechsel in der Tageslichtlänge gibt. Die Ergebnisse zeigen die Existenz einer jährlichen sensitiven Phase für lange Photoperioden zwischen Mitte November und März/April. Während dieser sensitiven Phase führt eine Exposition in eine lange Photoperiode (LD16:08) innerhalb von drei Wochen zum Gonadenwachstum. Die Experimente verdeutlichen außerdem, dass Europäische Feldhamster unter natürlichen Bedingungen lange Photoperioden nicht vor Mitte November wahrnehmen können. Dies ist ein wichtiger Unterschied zu anderen Hamster Spezies, bei denen das Gonadenwachstum zu jedem Zeitpunkt der Gonadenregression durch lange Photoperioden stimuliert werden kann. Weitere Experimente zeigten, dass eine Photoperiode von ca. 13h nötig ist, um das Gonadenwachstum zu stimulieren. Unter natürlichen Lichtbedingungen in Stuttgart (48°46'N) wird eine Photoperiode von 13h Anfang April erreicht, was gut zu dem Befund passt, dass der Großteil der Tiere, die unter LDnat gehalten wurden, Ende April voll entwickelte Gonaden hatte. Trotzdem zeigten diese Tiere ein relativ variables Timing der Gonadenentwicklung, was sich von Anfang Januar bis Ende April erstreckt. Dies ist wahrscheinlich das Resultat von zwei Prozessen: erstens einem endogenen Mechanismus, der die Wiederausbildung der Gonaden ohne jegliche Information induziert während die Tiere noch in ihren Winterbauen sind, und zweitens einem direkten stimulierenden Effekt von langen Photoperioden. Der zweite Teil dieser Dissertation untersucht die mögliche Rolle des circadianen Systems bei den Synchronisationsprozessen der circannualen Uhr. Diese Studie liefert die erste detaillierte Analyse der zeitlichen Beziehung zwischen saisonalen Zyklen von Reproduktion und Körpergewicht und saisonalen Veränderungen circadianer Parameter, nämlich der Laufradaktivität und der Exkretion von 6-Sulphatoxymelatonin (aMT6s), bei individuellen Tieren unter natürlichen Lichtbedingungen. Die Ergebnisse zeigen, dass die charakteristischen saisonalen Wechsel im Muster der Laufradaktivität und aMT6s-Exkretion um die Sommersonnenwende, also zwischen Mitte Mai und Mitte Juli, nur schwach mit Veränderungen im reproduktiven Status der Tiere zusammenhängen. Stattdessen korrelieren sie stark mit der Periode des jährlichen Zyklus, in der die Tiere sensitiv auf Kurztaginformationen reagieren (Saboureau et al., 1999) und sie könnten daher einen spezifischen Status des circadianen Schrittmachersystems widerspiegeln.
The distribution area of the European hamster (Cricetus cricetus) extends over the temperate zone of the Eurasian continent. Thus, the habitat is characterized by dramatic seasonal changes in climate, vegetation and food availability. In adaptation to these seasonal changes European hamsters exhibit pronounced seasonal variations in their physiology and behavior. The breeding season is restricted to spring and early summer, while the hard conditions of winter are avoided by hibernation. In addition body weight increases in the first half of year and decreases after the summer solstice. These seasonal changes have a strong endogenous component because they persist even under constant conditions (Canguilhem, 1989), i. e., they reflect most likely a true circannual rhythm. Finally, seasonal variations have been observed in two circadian functions, namely the daily activity pattern and the nocturnal melatonin secretion. During most parts of the year the activity rhythm is weak or even arhythmik, whereas high levels of activity and a precise activity pattern are observed only during two to three months around the summer solstice (Wollnik et al., 1991). The nightly melatonin secretion shows a complementary rhythm: Nocturnal melatonin excretion is high during short photoperiods and almost absent around the longest day (summer solstice) (Vivien-Roels et al., 1992). Usually, the period length of endogenous circannual rhythms differs from the annual cycle of the environment (that’s why they are called circa-annual). Thus the circannual clock has to be synchronized to the environmental oscillation at least once a year. It is well accepted that the annual changes in day length (photoperiod) provide the most important environmental signal to synchronize or entrain the circannual rhythm. This thesis deals with the question how the photoperiodic entrainment mechanism works in European hamsters. The synchronization of annual rhythms in the European hamster is the result of an interaction between seasonal changes in photoperiod and seasonal changes in the animal’s responsiveness to these photoperiods. One phase of sensitivity to short photoperiods was already discovered by (Saboureau et al., 1999) and lasts from mid-May to mid-July, when a decrease in day length induce gonadal regression and a decrease in body weight. In the first part of the present thesis the hypothesis of a sensitive phase to the opposite change of day length in winter was examined. The results demonstrated the existence of an annual phase of sensitivity to long photoperiods between mid-November and March/April. During this phase of sensitivity, exposure to a long photoperiod (LD16:8) induced gonadal development within 3 weeks. The experiments also demonstrated that under natural conditions European hamsters are not able to perceive long photoperiods (LD16:8) before mid-November. This is an important difference to other hamster species, in which regrowth of the gonads can be stimulated by exposure to long photoperiods at any stage of gonadal regression. Additional experiments indicated that a photoperiod of approximately 13 hours is necessary to stimulate gonadal development. Under natural lighting conditions in Stuttgart (48°46’ N), a photoperiod of 13 h is reached by early April, which fits well with the finding that the majority of animals kept under LDnat had well-developed gonads by the end of April. Nevertheless, these animals showed a rather variable timing of gonadal development, ranging from early January until late April. This is most likely the result of two processes, first an endogenous mechanism that induces gonadal recrudescence without any photoperiodic information while the animals are still in their hibernation burrows, and second a direct stimulatory effect of long photoperiods. The second part of the thesis examined a possible role of the circadian system in the synchronization process of the circannual clock. This study provides the first detailed analysis of the temporal relationship between seasonal cycles of reproduction and body weight and seasonal changes of two circadian parameters, i.e., locomotor activity and 6-sulphatoxymelatonin (aMT6s) excretion, in individual animals kept under natural light conditions. The results demonstrate a characteristic pattern of locomotor activity and aMT6s excretion observed around the summer solstice, i.e., from mid-May to mid-July. Locomotor activity was characterized by a high level of activity and an early onset of activity. Seasonal changes in aMT6s and in the pattern of locomotor activity were only loosely related to changes in the reproductive status of the animals. Instead, they correlated well with the period of the annual cycle during which the animals are sensitive to short days (Saboureau et al., 1999) and may thus reflect a specific state of the circadian pacemaker system.
